Recombinant Human TNFSF18 (GITRL) (Cat. No. 559204) (black circles) binds to immobilized Recombinant Human GITR (TNFRSF18) Fc Chimera (Cat. No. 764302). Ultra-LEAF™ purified anti-human CD357 (GITR) (clone W21071E) (purple squares) inhibits this binding in a dose-dependent manner, whereas the Ultra-LEAF™ purified rat IgG2a, κ isotype control (Cat. No. 400544) (gray triangles) does not have this effect. ND50 range: 0.3 - 2 µg/mL

Description

NFRSF18 is a type I transmembrane protein that was initially identified from dexamethasone-treated murine T cell hybridoma cells, and was designated GITR (for glucocorticoid-induced TNFR family-related gene). The human TNFRSF18 has 56% amino acid sequence identity to the mouse receptor. The human and mouse receptors contain three cysteine rich domains (CRD) in their extracellular region. TNFRSF18 expression is up-regulated in human peripheral mononuclear cells after stimulation with anti-CD3/CD28 monoclonal antibodies. TNFRSF18 interacts with TRAF1 (TNF receptor-associated factor 1), TRAF2, and TRAF3, and induces nuclear factor NFkB activation via TRAF2. TNFRSF18 is a key player in the differentiation of thymic Tregs (tTregs), and expansion of both tTregs and peripheral Tregs (pTregs). Human regulatory T cells (Tregs) constitutively expresses TNFRSF18. TNFRSF18 and its ligand are involved in the pathogenesis of autoimmune disease.  Soluble forms for TNFRSF18 and TNFSF18 were detected in the serum of patients with primary Sjögren's syndrome, and those levels correlated with disease severity. Other autoimmune and allergic diseases have been associated to TNFRSF18 /TNFSF18 interaction as demonstrated in disease models of gastritis, thyroiditis, experimental autoimmune encephalomyelitis, and allergic airway inflammation.

Application

Block - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.15 µg/mL human TNFSF18 (GITRL) to 2 µg/mL immobilized recombinant human GITR (TNFRSF18) Fc Chimera. ND50 range: 0.3-2 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Related FAQs

Do you guarantee that your antibodies are totally pathogen free?

BioLegend does not test for pathogens in-house aside from the GoInVivo™ product line. However, upon request, this can be tested on a custom basis with an outside, independent laboratory.

Does BioLegend test each Ultra-LEAF™ antibody by functional assay?

No, BioLegend does not test Ultra-LEAF™ antibodies by functional assays unless otherwise indicated. Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

Does BioLegend test each Ultra-LEAF™ antibody for potential pathogens?

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.

Have you tested this Ultra-LEAF™ antibody for in vivo or in vitro applications?

We don't test our antibodies for in vivo or in vitro applications unless otherwise indicated. Depending on the product, the TDS may describe literature supporting usage of a particular product for bioassay. It may be best to further consult the literature to find clone specific information.
