Total cell lysates (15 µg total protein) from PC-3 (negative control), HeLa (negative control), THP-1 and HEL cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-SHIP-1 Antibody, clone P1C1-A5, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

    THP-1 cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. The cells were then intracellularly stained with either (A) 2 µg/ml anti-mouse IgG1, κ or (B) 2 µg/ml SHIP-1 antibody (clone P1C1-A5) , (C) 0.5 µg/ml SHIP-1 antibody (clone P1C1-A5), or (D) 0.2 µg/ml SHIP-1 antibody (clone P1C1-A5), and incubated overnight at 4°C followed by 2 µg/ml (1:250) Alexa Fluor® 488 (Green) goat anti-mouse IgG for one hour at room temperature. Nuclei were counterstained with DAPI (Blue). The image was captured with a 60X objective. Exposure times: (A) 1/5, (B) 1/7.5, (C) 1/7.5, (D) 1/7.5.

Description

SH2-containing inositol phosphatase 1 (SHIP1) is a hematopoietic phosphatase that hydrolyzes key signaling lipid PI(3,4,5)P(3) to PI(3,4)P(2). SHIP-1 is located in the cytoplasm and is recruited to the cell surface upon receptor ligation. It plays critical roles in hematopoietic cell proliferation, activation, differentiation, and survival by regulating PI3K and Akt pathways. Aberrant regulation of PI3K signaling can induce autoimmune diseases and cancer formation.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 4.0 ug/ml dilution. For immunocytochemistry, the suggested use is 0.5 - 1ug/ml (1:500-1:1000 dilution). It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Specific to human SHIP-1 and cross reactive with mouse SHIP-1.
