Whole cell extracts (15 µg total protein) from SH-SY5Y, A549, Daudi, and HeLa (positive control) lysates, and NIH/3T3 lysates were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL (1:500 dilution) purified anti-TSG101 antibody (clone W19019A) overnight at 4°C. Description

Tumor Susceptibility Gene 101, TSG101, is a member of the ubiquitin E2 variant (UEV) subfamily of ubiquitin-conjugating enzymes. It is vital to transcriptional regulation, cytokinesis, tissue homeostasis, cell cycle regulation, and cell growth, differentiation, and proliferation. TSG101 is an essential component of the ESCRT-1 (endosomal sorting complexes required for transport) complex, which functions in multivesicular body biogenesis, endosomal trafficking, management of damaged proteins, recycling of membrane receptors, and cellular abscission. Within ESCRT-1, TSG101 binds to ubiquitinated proteins, aids in the formation of multivesicular bodies (MVBs), and recruits subsequent ESCRT components to the complex. Its role as part of ESCRT also makes TSG101 vital to cellular signal transduction, the recycling of receptor tyrosine kinases (RTKs), and Src-mediated migration. TSG101 is essential to the pathogenesis of several viruses - including HIV, influenza, and human papillomavirus - through its facilitation of interactions between the ESCRT complex and viral matrix proteins via the P(T/S)AP motif, as well as the budding and egress of enveloped viruses. Once predicted to behave as a tumor suppressor due to its role as a negative growth regulator, TSG101’s overexpression in colon, thyroid, breast, ovarian, and lung cancers suggests that TSG101 may actually function as an oncoprotein and play a role in cancer progression.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

ICC testing was conducted on HeLa siRNA control cells (positive control) and HeLa TSG101 siRNA knockdown cells (negative control) using three different fix/perm methods: MeOH fixation, PFA + Triton X-100, and PFA + MeOH. Staining was observed but there was no decrease in signal observed in the siRNA knockdown cells, indicating staining was not specific for TSG101. We therefore do not recommend this clone for the application.
