    The quantification of Human phospho-EGFR (Tyr1173) is demonstrated by treating A431 cells with or without the EGFR kinase inhibitor PD153035. A431 cells were incubated with or without recombinant human EGF (Cat. No. 585506) at 100 ng/mL after being pre-treated with or without the EGFR-kinase inhibitor PD153035 at either 20 nM or 100 nM. Cells were collected and then lysed with ice-cold lysis buffer. BCA assay was performed to measure total protein concentration. All lysate samples were diluted down to 10 µg/mL total protein in 1X ADB before being analyzed with the ELISA MAX™ Deluxe Set Human Phospho-EGFR (Tyr1173) assay. Then, the measured phospho-EGFR (Tyr1173) was normalized to the total protein concentration.

Description

EGFR (Epidermal Growth Factor Receptor) is a member of the ErbB/HER family of cell surface receptors which comprises of four closely related tyrosine kinase receptors: EGFR/HER1, HER2, HER3, and HER4. Binding of a ligand to the extracellular domain of EGFR results in receptor dimerization and autophosphorylation on tyrosine residues with the major phosphorylation sites being tyrosine 1068 (Tyr1068), Tyr1148, and Tyr1173. Phosphorylation of Tyr1173 leads to interaction with Shc and phospholipase Cγ (PLCγ), which are involved in the activation of MAPK signaling pathway. EGFR activation triggers intracellular signaling that results in increased proliferation and a number of processes related to cell survival. ErbB receptors are overexpressed or mutated in many cancers. It was reported that deficient signaling of the EGFR can result in degenerative diseases such as Alzheimer’s disease.

No, ELISA MAX™ Deluxe Sets do not come with plates. Coating Buffer and Assay Diluent (for blocking and dilutions) are included in these sets. Plates can be ordered separately (Cat No. 423501), and instructions for coating the plates are in the sets’ product manuals.
