Total cell lysates (15 µg protein) from P116, Jurkat and EL-4 were resolved by 4-20% Tris-Glycine gel electrophoresis, transferred to nitrocellulose, and probed with purified anti-ZAP70 (clone A15114B) antibody. Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin (clone 2F1-1) antibody was used as a loading control.

Total cell lysates (15 µg protein) from Ramos (lane 1), Jurkat (lane 2), EL4 (lane 3) and A20 (lane 4) cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 1 µg/mL (1:500 dilution) of purified anti-ZAP70 antibody (clone A15114B) (upper blot). Proteins were visualized using chemiluminescence detection after incubation with a 1:3000 dilution of HRP conjugated anti-mouse-IgG secondary antibody for anti-ZAP70 antibody (upper blot) or a 1:5000 dilution of Direct-Blot™ HRP anti-β-Actin (clone 2F1-1, lower blot) antibody. Description

ZAP70 is a 70 kD protein tyrosine kinase associated with the zeta chain of the T cell receptor. It is expressed in T cells and NK cells and has been shown to be involved in T cell signaling. Defects in ZAP70 have been linked to selective T cell defects. The ZAP70 kinase undergoes multiple phosphorylation events after T cell receptor engagement and interacts with a number of proteins involved in signal transduction. Recently, ZAP70 has been identified as an important prognostic marker in B-cell chronic lymphocytic leukemia (B-CLL).

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. For intracellular flow cytometric staining, the suggested use of this antibody is ≤ 0.06 µg per million cells in 100 µl volume. It is recommended that the reagent be titrated for optimal performance for each application.
