Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 0.25 µg/mL (1:2000 dilution) of purified anti-TIGAR antibody (clone W18015A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker. Cell lysates were loaded in ascending order of predicted TIGAR expression; predicted expression data was obtained from Human Protein Atlas.

Description

Tp53-induced glycolysis and apoptosis regulator (TIGAR) is a negative regulator of glycolysis upregulated by p53 in response to cellular stresses and it protects cells against reactive oxygen species (ROS)-induced autophagy. TIGAR decreases the rate of glycolysis by reducing the level of fructose-2, 6-bisphosphate, which is an activator of phosphofructokinase-1 (PFK1), a rate-limiting enzyme in glycolysis. Additionally, TIGAR promotes the pentose phosphate pathway and provides NADPH for glutathione reductase to generate reduced glutathione (GSH). GSH lowers the intracellular level of ROS and prevents DNA damage, thereby promoting cell survival.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone does not recognize mouse TIGAR.

The clone produced incorrect staining in ICC testing.

The clone could not pull down TIGAR in IP testing.
