Whole cell extracts (15 µg total protein) of the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.5 µg/mL purified anti-TIF1γ/TRIM33 (clone 3H7-C8-B9) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: Molecular weight marker

ICC staining of purified anti-TIF1γ/TRIM33 (clone 3H7-C8-B9) on MCF7 cells. Cells were fixed with 4% paraformaldehyde, permeabilized with 0.5% Triton X, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 2.5 µg/mL of anti-TIF1γ/TRIM33 (clone 3H7-C8-B9) overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) (panel A) for 1 hour at room temperature. Nuclei were counterstained with DAPI (panel B) (Cat. No. 422801), and images were captured with 60X objective. Overlay image (panel C). Description

TIF1γ/TRIM33, belonging to the tripartite motif-containing (TIF) protein family, is a multifunctional protein that regulates hematopoiesis, DNA repair, mitosis, and transcription elongation. Just as with other TIF/TRIM family members, TIF1γ/TRIM33 contains a conservative RBCC/TRIM motif at the N terminus and a PHD/ bromodomain at the C terminus.  TIF1γ/TRIM33 suppresses the TGF-beta/BMP signaling pathway via monoubiquitylation of SMAD4 or the binding of phosphorylated SMAD2/3. Through its E3 ubiquitin ligase function, TIF1γ/TRIM33 decreases nuclear beta-catenin expression and suppresses tumorigenesis and tumor cell proliferation. TIF1γ/TRIM33’s tumor suppressive properties have been observed in hepatocellular carcinoma, chronic myelomonocytic leukemia, and pancreatic cancer. It has also been observed to reduce the stability of HIV-1 viral integrase to restrict HIV-1 infection.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product is not recommended for use in IHC-P.

For immunocytochemistry staining, the suggested fixation method is 4% paraformaldehyde.
