Whole cell extracts (15 µg total protein) from indicated cell lines or tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 µg/mL of purified anti-Sun2 (clone 3.1E) overnight at 4°C.     NIH-3T3 cells were fixed and permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 10 µg/mL of purified anti-Sun2 (clone 3.1E), followed by incubation with Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature (panels A and B). Nuclei were counterstained with DAPI (Cat. No. 422801) (panel A). The images were captured using a Revvity Operetta CLS™ High Content Analysis System with a 63X water objective. Scale bar: 50 µm

    IHC staining of purified anti-Sun2 (clone 3.1E) on formalin-fixed paraffin-embedded mouse brain. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated with 10 µg/mL of purified anti-Sun2 (clone 3.1E) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. Description

Sun2 is a transmembrane protein that localizes to the inner nuclear membrane and interacts with other nuclear proteins to form the LINC (Linker of Nucleoskeleton and Cytoskeleton) complex. This complex bridges the nuclear envelope, connecting the nuclear lamina to the cytoskeleton, thereby providing mechanical support and stability to the nucleus. Sun2 also plays a vital role in nuclear migration, chromosome organization, and gene regulation. Additionally, Sun2 is involved in maintaining nuclear envelope integrity during cell division and DNA repair, and has been suggested to act as a tumor suppressor by protecting against cancer-inducing DNA damage. In lung cancer, Sun2 has been shown to inhibit the Warburg effect via Glut1 and LDHA.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For IHC-P, antigen retrieval using Citrate buffer, pH 6.0 (Cat. No. 420901) is recommended.

For ICC, we recommend fixation/permeabilization with 100% ice-cold methanol for 10 minutes. Other fixation/permeabilization methods have not been tested.
