Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL of purified anti-RBX1 (clone W19401A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat.     ICC staining of purified anti-RBX1 (clone W19401A) on HeLa cells (panel B) or purified rat IgG2b, κ isotype control (panel A) (Cat. No. 400602). The cells were fixed and permeabilized with 4% paraformaldehyde, permeabilized with 0.5% Triton X, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 5.0 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801), and the image was captured with a 60X objective. Scale bar = 50 µm

Description

RBX1 is a RING (Really Interesting New Gene) finger-like domain-containing protein. RBX1 is an E3 ubiquitin ligase component of multiple cullin-RING-based E3 ubiquitin-protein ligase (CRLs) complexes that mediate ubiquitination and subsequent proteasomal degradation of target proteins, including those involved in cell cycle progression, signal transduction, and transcription. RBX1 also promotes the neddylation of CUL1, CUL2, CUL4 and CUL4 via its interaction with UBE2M.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product is not recommended for IHC-P.
