Whole tissue membrane cell extracts (15 µg total protein) from human and rat brain were heated at 37°C for 30 minutes, resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified anti-Iduna (RNF146) antibody (clone N201/35) overnight at 4 °C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular weight marker.

Description

Ring finger protein 146 (RNF146), also known as Iduna, is a poly-ADP-ribose (PAR)-dependent E3 ubiquitin-protein ligase that promotes the degradation of PARsylated proteins. The ability of RNF146 to bind to PAR and thus inhibit PAR dependent cell death, or parthanatos, contributes to its role in DNA repair and cell survival and overall neuroprotective qualities. RNF146 has been identified as an NMDA receptor-induced survival gene that protects against NMDA receptor mediated excitotoxicity and ischemia-induced neuronal injury. RNF146 is a positive regulator of the Wnt/ β-catenin signaling pathway through the ubiquitylation of AXIN following its degradation by tankyrase and has been implicated in embryonic and postnatal osteoblast development, bone remodeling, and energy metabolism via coordination of β-catenin signaling and Hippo pathways within osteoblasts.
