  • Human_FOLR1_1_CF
Biotinylated Recombinant Human FOLR1 binds to immobilized FA-BSA (folic acid-BSA conjugate) in a dose-dependent manner. The ED50 for this effect is 0.5-3.0 ng/mL.
  • Human_FOLR1_2_CF
Stability Testing for Recombinant Human FOLR1. Biotinylated Recombinant Human FOLR1 was aliquoted in PBS, pH 7.0 at 0.2 mg/mL and one aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability to bind to FA-BSA (folic acid-BSA conjugate) in a dose-dependent manner. The ED50 for this effect is 0.5-3.0 ng/mL

Description

FOLR1, also known as folate receptor alpha, is a glycosylphosphatidylinositol (GPI)-anchored cell-surface glycoprotein encoded by the FOLR1 gene. FOLR1 binds with high affinity to folic acid and its derivatives and mediates cellular processes, including cell division, proliferation, and tissue growth via signaling cascades and components of the folate cycle. Each FOLR only binds one folate and brings the folate into cells via endocytosis of the FOLR-folate complex. Human FOLR1 shares 83% amino acid sequence homology with mouse and rat FOLR1. FOLR1, the most extensively studied isoform, has limited tissue expression but is overexpressed in several tumors of epithelial origin including ovarian cancer, non-small cell lung adenocarcinoma, breast cancer, renal cancer and endometrial cancer. The overexpression of FOLR1 is often associated with increased cancer progression and poor patient prognosis. It plays a critical role during embryogenesis.

Product Details

Source
Human FOLR1, amino acid (Arg25-Met233) (Accession: # P15328) with a C-terminal 8His and Avi tag, was expressed in CHO cells.
Molecular Mass
The 246 amino acid recombinant protein has a predicted molecular mass of approximately 28.4 kD. The DTT-reduced and non-reduced protein migrates at approximately 35-45 kD and 35-40 kD respectively as determined by SDS-PAGE. The predicted N-terminal amino acid is Arg.
Purity
> 95%, as determined by Coomassie stained SDS-PAGE.
Formulation
0.22 µm filtered protein solution is in PBS
Endotoxin Level
Less than 0.1 EU per µg cytokine as determined by the LAL method.
Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at - 20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
Biotinylated Recombinant Human FOLR1 binds to immobilized FA-BSA (folic acid BSA conjugate). The EC50 of this effect is 0.5 – 3.0 ng/mL.

Application Notes

BioLegend carrier-free recombinant proteins provided in liquid format are shipped on blue-ice. Our comparison testing data indicates that when handled and stored as recommended, the liquid format has equal or better stability and shelf-life compared to commercially available lyophilized proteins after reconstitution. Our liquid proteins are validated in-house to maintain activity after shipping on blue ice and are backed by our 100% satisfaction guarantee.
