IHC staining of purified anti-PGR (clone BSB-2) on formalin-fixed paraffin-embedded human uterus tissue. Following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, pH 9.0), the tissue was incubated without (panel A) and with (panel B) 5 µg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat.     MCF-7 cells were fixed with 4% Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then incubated with 1.25 µg/mL of either purified mouse IgG1, κ isotype control (Cat. No. 400102) (Panel A) or purified anti-PGR (clone BSB-2) (Panel B), followed by incubation with Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature.     Whole cell extracts (15 µg total protein) from the indicated cell lines and tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and incubated with 1 µg/mL of purified anti-PGR (clone BSB-2) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

PGR (Progesterone receptor) is a nuclear receptor, a type of receptor protein that directly interacts with DNA to affect gene expression upon ligand binding. PGR is activated by the hormone progesterone, a steroid and sex hormone. Widely expressed in female reproductive tissues, activation of PGR is critical for the establishment and maintenance of pregnancy and in the development of the mammary glands. PGR is also expressed in the central nervous system, modulating innervation by dopaminergic neurons. Overexpression of PGR is often found in breast, ovarian and uterine cancers, and PGR is one of the three major receptors assessed when classifying breast tumors.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration of 1.25 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product does not recognize mouse progesterone receptor in Western blot.

This product only recognizes PGR Isoform B.

For immunocytochemistry, fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with 0.5% Triton-X is recommended. Fixation with Fixation Buffer followed by permeabilization with 100% methanol, or fixation with 100% methanol are not recommended.
