Total cell lysates (15 µg total protein) from HEL (negative control, Lane 1), HeLa (reduced expression control, Lane 2), and Daudi cells (Lane 3) were resolved by 4-20% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-FOXO1 Antibody, clone 2F8B08, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse-IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH Antibody (Cat.     IHC staining using purified anti-FOXO1 (clone 2F8B08) on formalin-fixed paraffin-embedded human cerebellum tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated with 10 μg/mL of antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG antibody (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. Description

FOXO1 is a transcription factor, belonging to the O subfamily of forkhead box-containing proteins. The forkhead proteins are substrates of Akt/PKB and glucocorticoid-induced kinases that mediate insulin signaling. FOXO1 binds to the conserved insulin-responsive elements and acts as a transcriptional activator. The FOXO1 target genes are involved in cell metabolism, growth, and differentiation. Upon insulin stimulation, FOXO1 proteins are phosphorylated by AKT/PKB through the PI3K pathway, resulting in the translocation of FOXO1 proteins from the nucleus to the cytoplasm. The expression of FOXO1 target genes are then inhibited by FOXO1. FOXO1 has been reported to regulate maturation and class-switch recombination of B cells, as well as participating in regulatory T cell function. FOXO1-deficient mice are embryonic lethal, suggesting that FOXO1 has crucial functions in development which cannot be compensated by other FoxO family members.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. For immunocytochemistry, a concentration of 5.0 μg/ml is recommended. For immunoprecipitation, the suggested use of this reagent is 5.0 - 20 μg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 8.0 - 10. µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
