Whole tissue membrane extracts (15 µg total protein) from rat and mouse brain were heated at 37°C for 30 minutes, resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified anti-MAP3K12 antibody (clone N377/20) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. Description

Mitogen-activated protein kinase kinase kinase 12 (MAP3K12), also known as dual leucine zipper kinase (DLK), is a serine/threonine-protein kinase and part of the non-canonical MAPK signaling pathway. Primarily expressed in neurons at the synaptic terminals, MAP3K12 is a critical regulator of signaling cascades affecting neuronal migration and axon development as well as responding to peripheral nerve injury including transcriptional repression, axon regeneration, and neuronal apoptosis through its activation of transcription factors STAT3 and c-Jun. Its neuroprotective role in responding to injury makes MAP3K12 a promising therapeutic target in the study of neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease, amyotrophic lateral sclerosis (ALS), and optic neuropathy. Emerging evidence suggests a role for MAP3K12 in the proliferation and mass regulation of pancreatic beta cells via activation of c-Jun N-terminal kinase (JNK) signaling pathway and may be a target of interest in the study of the pathogenesis of insulin resistance.
