Whole cell extracts (15 µg total protein) from the indicated cell lines and tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL of purified anti-IBA1 (clone A19290E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat.     IHC staining of purified anti-IBA1 (clone A19290E) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using 1X Tris-EDTA, pH 9.0, the tissue was incubated without (panel A) and with (panel B) 10 µg/mL of purified anti-IBA1 (clone A19290E) incubated overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 Goat anti-mouse IgG (minimal x-reactivity) (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 63X objective. Scale bar: 20 µm

    THP-1 cells (positive control, filled histogram) and HeLa cells (low expression control, open histogram) were fixed and permeabilized using Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) and intracellularly stained with purified anti-IBA1 (clone A19290E) or purified mouse IgG2a, κ isotype control (open histogram, dashed line) (Cat. No. 400202), followed by incubation of PE Goat anti-mouse IgG (minimal x-reactivity) (Cat. No. 405307) for 30 minutes in the dark.

Description

As a cytoplasmic scaffold protein, IBA1 contains Ca2+ binding EF-hand and PDZ interaction domains, which are important for mediating intracellular signaling complexes. IBA1, also known as AIF-1, was first observed in rodent microglial cells and has been shown to stain cells in both normal and pathological lesions in rodents. In addition to the central nervous system, IBA1 has also been shown to stain in macrophage subpopulations. In macrophages, expression of IBA1 promotes pro-inflammatory responses. In microglial cells, IBA1 is important in the modulation of synaptic activities and is upregulated in response to nerve damage.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

This clone is not mouse or rat reactive.

This clone is not suitable for ICC.
