IHC staining using purified anti-SDHB (clone W19244A) (panel B) or secondary-only control (panel A) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-EDTA buffer the tissue was incubated with 10 μg/mL of antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-rat IgG (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale bar = 50 µm

    Whole cell extracts (15 µg total protein) from indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-SDHB (clone W19244A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

    PC-3 cells were fixed with ice cold methanol for 10 minutes and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with purified rat IgG2a, κ isotype control (Cat. No. 400502) (Panel A) or 5.0 µg/mL purified anti-SDHB (clone W19244A) (Panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) at 2.0 µg/mL. Description

SDHB encodes one of the four proteins that form the SDH (succinate dehydrogenase) enzyme complex in the mitochondrial-respiratory chain, and plays an important role in the mitochondria. The SDH complex functions to couple the conversion of succinate to fumarate in the mitochondrial matrix as part of the Krebs cycle with the reduction of ubiquitin to ubiquinol as part of the ETC (electron transport chain). Within the complex itself SDHB provides an attachment site for released electrons to be transferred to the oxidative phosphorylation pathway. Sporadic and familial mutations in this gene result in paragangliomas, pheochromocytoma, and gastrointestinal stromal tumors.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone W19244A is suitable for ICC using methanol only as the fixation/permeabilization method. Alternate methods such as fixation with PFA followed by permeabilization with either methanol or Triton-X-100 resulted in staining in the wrong localization.
