Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.5 µg/mL (1:1000 dilution) of purified anti-PGK1 (clone W18295A) overnight at 4°C.     IHC staining of purified anti-PGK1 (clone W18295A) on formalin-fixed paraffin-embedded mouse eye tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1x (Cat. No. 928502), the tissue was incubated with 5.0 µg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, DAB) (Cat.     ICC staining of purified anti-PGK1 (clone W18295A) on HeLa cells. The cells were fixed and permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with either purified rat IgG2a, λ isotype control (Cat. No. 402301) (left panel) or purified anti-PGK1 (clone W18295A) (right panel) at 1.0 µg/mL overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (4',6-Diamidino-2-Phenylindole, Dilactate) (Cat. No. 422801), and the image was captured with a 40X objective. Description

Phosphoglycerate kinase 1 (PGK1) is one of two enzymes, along with its isozyme PGK2, which catalyzes the conversion of 1,3-diphosphoglycerate to 3-phosphoglycerate, and transfers the phosphate group to ADP to produce ATP. This step is notable as the “break even” point of glycolysis where ATP expenditure matches ATP output. Additionally, PGK1 functions as a protein kinase, and is also involved in transcription initiation as a cofactor for DNA polymerase alpha. The gene encoding PGK1 is on the X chromosome, and mutations in PGK1 are associated with various cancers, and X-linked recessive variants of anemia, mental disorders, and myopathy.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1 µg/mL. For immunohistochemistry, a concentration range of 1 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
