Total cell lysates (15 µg total protein) from Jurkat, MCF-7, HeLa, and NIH3T3 were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-NFATC3 Antibody, clone W15221A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

Nuclear factor of activated T cells C3 (NFATC3), also known as NFAT4 or NFATx, is a member of the NFAT family of transcription factors. Several NFATC3 variants generated by alternative splicing were identified. NFATC3 plays an important role in the regulation of T cell activation-related gene expression, especially in the induction of IL-2.  NFATC3 is expressed in immune cells, such as double positive (DP) thymocytes, mature CD4+ T cells, and mast cells. It is also found in non-immune cells such as smooth muscle and Schwann cells. In the resting T cells, NFATC3 is located in the cytoplasm and is phosphorylated. Upon T cell activation by TCR stimulation, the increase of intracellular Ca2+ induces NFATC3 dephosphorylation via calcineurin. Dephosphorylated NFATC3 exposes the nuclear localization signal and is subsequently translocated from the cytosol to the nucleus. Through the highly conserved Rel-like binding domain, NFATC3 associates with specific consensus DNA sequences and regulates gene expression.

Application

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone W15221A may react with isoforms 1-4 (MW 112.6~115.6). NFATs are heavily phosphorylated proteins, so the observed MWs of NFATC3 can be higher than predicted.

Antigen Details

Structure
There are 6 reported isoforms of NFATC3 (MWs: 115.1, 115.2, 115.6, 112.6, 77.2, and 80.6 kD). The protein contains a RHD (Rel-like) domain responsible for DNA binding.
Distribution

Cytoplasm and nucleus.

Function
NFATC3 is a component of activated T cells DNA-binding transcription complex, regulating gene expression in immature thymocytes and T cells.
Interaction
Forms a multicomponent complex with NFATC1 and NFATC2. Interacts with Raf-1.
Biology Area
Cell Biology, Transcription Factors
