    Human paraffin-embedded colon cancer tissue slices were prepared with a standard protocol of deparaffinization and rehydration. Antigen retrieval was done with Sodium Citrate H.I.E.R. 1X at 95°C for 40 minutes. Tissue was washed with PBS/0.05% Tween 20 twice for five minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the tissue was stained with 10 µg/mL of anti-human CD44v9 (clone RV3) purified at 4°C overnight, followed by 2.5 µg/ml of anti-rat IgG (clone Poly4054) Alexa Fluor® 647 (red) for 2 hours at room temperature. Nuclei were counterstained with DAPI (green). The image was captured with a 10X objective.

Description

CD44 isoform 9, also known as CD44v9, is a CD44 isofom consisting of exons 1-5, 13 (v9) and 15 to 19, product of alternative splicing. CD44v9 is expressed by cancer stem cells, epithelial-type carcinomas, subsets of monocytes and subsets of lymphocytes; have a role in tumor initiation, maintenance and metastasis of cancer cells. CD44v9 interacts with the glutamate-cystine transporter xCT, increasing reduced glutathione and contributing to the ROS resistance of cancer cells, and is a potential predictive marker for recurrence of some cancers and a potential therapeutic target.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤1.0 µg per million cells in 100 µl volume. For immunohistochemistry, a concentration range of 5.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
