Recombinant human EGF (Cat. No. 585506) (black circles) inhibits proliferation in human epithelial A431 cells. Ultra-LEAF™ purified anti-human EGFR (clone W21030C) (purple squares) blocks the inhibitory effect of recombinant human EGF, at 10 ng/mL, in the proliferation of human epithelial A431 cells in a dose-dependent manner; whereas the Ultra-LEAF™ purified rat IgG2b, κ isotype control (Cat. No. 400644) (gray triangles) does not have effect. ND50 range: 0.15 - 1.5 µg/mL.

Epidermal Growth Factor Receptor (EGFR) is a receptor tyrosine kinase that links extracellular mitogenic ligand binding to complex downstream signaling cascades. Initial ligand binding results in receptor oligomerization and autophosphorylation of multiple tyrosine residues within cytosolic domains of the protein. These phosphorylation events stabilize the EGFR kinase activation loop and lead to the recruitment of adaptor proteins and other downstream effectors. EGFR stimulation leads to activation of RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCγ-PKC and STAT signaling cascades, and constitutive activation of the receptor promotes tumorigenesis in multiple cancers.

ND50 range: 0.15 - 1.5 μg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
