Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-SMAD2 (clone 6H5-E3-C11) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker

Description

SMAD2 is a potent regulator of different cellular processes such as apoptosis, differentiation, and cell proliferation. SMAD2 is associated with SARA protein and the TGF-B receptor. SMAD2 gets phosphorylated by the TGF-B receptor in response to TGF-B signal, which induces its dissociation from SARA and association with SMAD4 followed by translocation into the nucleus. In the nucleus, the SMAD2/SMAD4 protein participates in gene regulation by forming a transcription repressor complex in association with other cofactors.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Since SMAD2 shares a high percentage of identity with SMAD3, this clone recognizes both SMAD2 and SMAD3, which can be resolved by Western Blot.

Antigen Details

Structure
SMAD2 is a 467 amino acid protein with a predicted molecular weight of 52 kD. SMAD2 is very similar in sequence and structure to SMAD3.
Distribution

Non-phosphorylated SMAD2 is present in the cellular cytoplasm close to the TGF-B receptor. Phosphorylated SMAD2 is present in the cellular nucleus bind to target promotor sequences. 

Function
The function of SMAD2 is mainly as a gene expression regulator. SMAD2 acts as a sequence specific transcriptional repressor.
Interaction
Non-phosphorylated SMAD2 interacts with SARA (SMAD anchor for receptor activation) protein. Phosphorylated SMAD2 interacts with SMAD4 and other members of the transcription repressor complex.

SMAD2 is part of the TGF-B signaling pathway but it is not a ligand/receptor protein
