Whole cell extracts (15 µg total protein) from A549 (negative control), HeLa, U87-MG, and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) purified anti-Hexokinase II antibody (clone W19163B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker.

Description

Hexokinase enzymes are evolutionarily conserved proteins that play a critical role in glycolysis by catalyzing the conversion of glucose to the intermediate glucose-6-phosphate. In humans, four hexokinase isozymes exist that differ in their tissue distribution and affinity for glucose. The hexokinase II isozyme is ubiquitously expressed and is the most highly regulated member of the hexokinase family. In addition to its cytosolic localization, hexokinase II also associates with the mitochondrial outer membrane, resulting in efficient substrate channeling of ATP to the enzyme which fuels cell proliferation. Hexokinase II association with the outer mitochondrial membrane protein VDAC disrupts the ability of the pro-apoptotic protein Bax to induce mitochondrial dysfunction and cell death through cytochrome C release. Many cancer cells show elevated expression of hexokinase II, which allows efficient funneling of glucose to support anaerobic glycolysis, as well as increased survival through inhibition of Bax-mediated apoptotic death.

Application

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

-This clone was tested for ICC on HeLa cells fixed and permeabilized with 4% PFA plus Trixon X-100, 4% PFA plus methanol, or methanol. The former two methods resulted in nuclear staining, inconsistent with the published cytosolic and mitochondrial outer membrane-associated localization of hekokinase II. The latter method (methanol only) resulted in hexokinase II staining, but the signal observed did not meet our sensitivity criteria. We therefore do not recommend this clone for the application.

-This clone failed to immunoprecipitate hexokinase II from HeLa extracts. We therefore do not recommend this clone for the application.
