Whole tissue membrane extracts (15 µg total protein) from human, rat, and mouse brain tissue were heated at 37°C for 30 minutes, resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified anti-Cavβ4 antibody (clone N10/7) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. Description

Voltage-dependent L-type calcium channel subunit beta-4 (Cavß4) is a member of the membrane-associated guanylate kinase (GK) protein family and a cytoplasmic subunit of voltage-gated calcium channels. Expressed predominantly in the cerebellum and kidney, it is responsible for calcium entry into nerve termini and subsequent neurotransmitter release via its association with membrane-trafficking protein synaptotagmin 1 (Syt 1). It regulates synaptic transmission, increases the readily releasable pool of synaptic vesicles, and plays a role in synaptic vesicle plasticity and maturation through its association with Cavß1, RIM1, and actin filaments (F-actin). Cavß4 has been linked to psychiatric and anxiety disorders including major depressive disorder (MDD) and schizophrenia and is a potential therapeutic target in their study and treatment.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
