Western blot of anti-Alpha β Crystallin antibody (clone 1329CT523.140.120). Total tissue lysates (15 µg total protein) from human skin (negative control, Lane 1), human brain (positive control, Lane 2), mouse brain (positive control, Lane 3), and rat brain (positive control, Lane 4) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.5 μg/mL (1:1,000) purified anti-Alpha β Crystallin antibody overnight at 4 °C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3,000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25,000 dilution (lower). Lane M: Molecular weight ladder.

    IHC staining of purified anti-Alpha β Crystallin antibody (clone 1329CT523.140.120) on formalin-fixed paraffin-embedded human heart (A, positive control) and tonsil (B, negative control) tissues. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissues were incubated with 5 µg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, DAB, Cat. No. 929501) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The images were captured with a 40X objective. Scale bar: 50 μm.

Description

Alpha B Crystallin (CryAB) is a member of the small heat shock protein (HSP20) family.  Alpha crystallins are composed of two gene products: alpha-A and alpha-B, for acidic and basic, respectively. Alpha-A is preferentially restricted to the lens and alpha-B is expressed widely in many tissues and organs. CryAB can be induced by heat shock, ischemia and oxidation. The protein is known to protect cells from stress damage. It may also play an important role in the Golgi reorganization during cell division. CryAB is also known to be associated with a number of neurodegenerative diseases such as Alzheimer’s disease and has been implicated in breast tumors of patients with shorter survival, suggesting that it may be a useful marker in identifying aggressive breast cancer.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2.5 - 10.0 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone recognizes alpha β crystallin proteins at 21 kD in human, mouse, and rat brain lysates. An extra band at 15 kD is observed in the human brain lysates.
