Preserving cells is critical at particular stages in the workflow, so as to lengthen product shelf-life and allow for transport and storage to and from sites. First, the original tissue isolate can be stored and stabilized in a cryopreservation solution until processing begins. Second, after cell expansion, cryopreservation ensures that important qualities of processed cells are maintained for downstream applications.

After a 9-day expansion protocol using anti-human CD28, anti-human CD3, and IL-2, activated and expanded T cells were cryopreserved and thawed. (A) Viability and (B) cell recovery were determined at thaw. (C) Expansion ability was assessed after a 6-day (144 hours) culture period. The expansion fold was calculated by dividing the number of viable cells at day 6 by the number of viable cells at 2h post-thaw. (D) Flow cytometry was used to quantify CD4+ and CD8+ T cell populations at thaw, after the above 9-day expansion protocol.

Multipotent mesenchymal stem cells (MSCs) can differentiate into a variety of cell types, including osteoblasts, chondrocytes, myocytes, and adipocytes. Their clinical value extends to applications such as tissue engineering, wound healing, and other conditions in which regenerated cells can improve or resolve disease. Cell-Vive™ CD DMSO-Free Cryopreservation Solution, GMP preserves human MSC viability and phenotype, post-cryopreservation, at similar or higher levels to that of a commercially available 10% DMSO solution (Competitor).

After reaching 80% confluency, Human MSCs were detached and stored in liquid nitrogen for two days before thawing. (A) Viability at thaw was determined and (B) cell expansion demonstrated through three passages.

 

Cell-Vive™ CD DMSO-Free Cryopreservation Solution, GMP preserves the morphology of human MSCs similar to a commercially available 10% DMSO solution (competitor).

Human bone marrow-derived MSCs were frozen at 500,000 cells/mL in (A) Cell-Vive™ CD DMSO-Free Cryopreservation Solution, GMP or (B) a 10% DMSO solution from a competing supplier. Cells were stored in liquid nitrogen for two days, then thawed and plated at 5,000 cells/cm2. Attachment and morphology were observed four days after thawing. Images were taken at 4X magnification.

 

Cell-Vive™ CD DMSO-Free Cryopreservation Solution, GMP preserves the differentiation capacity of human Mesenchymal Stem Cells.

 

 

Human MSCs were stored in liquid nitrogen in the presence of Cell-Vive™ CD DMSO-Free Cryopreservation Solution, GMP for two days. MSCs were thawed and differentiated into (A) osteocytes for nineteen days or (B) adipocytes for twenty-one days with respective differentiation media (left) or cell culture media as a control (right). Osteocytes were stained with Alizarin Red and adipocytes with Oil Red O.
