Western blot of purified anti-CAPNS1 antibody (Poly28643). Lane 1: Molecular weight marker; Lane 2: 40 µg of human brain membrane lysate; Lane 3: 40 µg of mouse brain membrane lysate; Lane 4: 40 µg of rat brain membrane lysate. The blot was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP Donkey anti-rabbit IgG Antibody (Cat. No. 406401). Enhanced chemiluminescence was used as the detection system. -
IHC staining of purified anti-CAPNS1 antibody (Poly28643) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 5 µg/mL of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm -

Calpain Small Subunit 1 (CAPNS1) is a member of calpain small subunit family, which is a small regulatory subunit of calpains. Calpains are calcium-dependent cysteine-type proteinases that are involved in many cellular processes including cell proliferation, migration, adhesion and apoptosis. They are also associated with neurodegeneration, including myotonic dystrophy. Among its related pathways are apoptosis modulation and integrin pathway.

-
Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2.0 - 10 µg/mL in human and 5.0 - 10 µg/mL for mouse and rat is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
