Worked Great! Used to induce NF-kB Activity in 293T cells (Recombinant Human TNF-α)
Overall: Product Quality: Ease of Use:
The protein worked great as a positive control for all my experiments to induce NF-kB activity, which was measured with a luciferase reporter gene as the readout.
Experimental Design

Application: Induction of NF-kB as a positive control
Cells used: HEK293T
Brief Protocol: HEK293T cells were plated at 6x10^5 cells/well in a 6-well plate. The next day, the cells were transfected with NF-kB luciferase reporter plasmid and a house-keeping Renilla reporter plasmid as an internal control. 24-hrs post transfection, cells were treated with the TNF-a at 50 ng/ml for 2 hours. Cells were then harvested for lysis and detection of luciferase activity.
Results Summary: Great fold inductions were observed when compared to the experimental samples and the negative control.
Additional Notes: Works great for tissue culture and with no observable cytotoxicity.
