EpCAM is Not Detected on SUM159 Cells

EpCAM is not expressed on the cell surface of SUM159 cells.
Experimental Design

Application: Flow cytometry
Cells used: SUM159
Brief Protocol: Cells were trypsinized, neutralized with DMEM 10%, and re-suspended in ice-cold PBS with 2% IFS at 1x10^6 cells per 100 uL. The cells were incubated on ice with 1:100 antibody for 30’, washed with 9 times the original volume of ice-cold PBS- with 2% IFS, and re-suspended in 500 uL ice-cold PBS- with 2% IFS.
Results Summary: EpCAM is not expressed by SUM159 cells.

Technical Service Notes

Trypsinization can affect epitopes since the process is enzyme-based. This can make it difficult for antibodies to find their target. This may explain the negative result. As such, we would suggest non-enzymatic or gentler methods to test and see if this result is consistent. Pacific Blue is also a very dim fluor, so a brighter one may be needed.
