Toll-like receptor 9 (TLR9) is important in innate immune signaling as it recognizes unmethylated CpG sequences present in intracellular bacteria and DNA viruses.  Expressed primarily on B cells and plasmacytoid dendritic cells (pDCs) (1), TLR9 signals through the MyD88 pathway leading to inflammatory cytokine production, particularly type I interferons.  Despite its importance in innate immunity, there was a lack of specific reagents for staining TLR9 in human samples.  We’ve released a new clone, S16013D, which demonstrates bright and specific staining on PBMCs.

We stained human PBMCs with clone S16013D and other commercially available TLR-9 antibodies.  While the antibody from competitor 1 non-specifically binds to all PBMC populations and the competitor 2 antibody gives little staining over background, clone S16013D demonstrates clear and specific staining on only B cells and pDCs.
