IHC staining of Purified anti-VWF (clone A21125D/G) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0), the tissue was incubated with 5 µg/mL of Purified anti-VWF (clone A21125D/G) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured using a Revvity Operetta CLS™ High Content Analysis System with a 40X objective. Scale bar: 50 µm

    HeLa cells (negative cell line, panel A) and HUVEC cells (positive cell line, panel B) were treated with Brefeldin A (Cat. No. 420601) for 4 hours and fixed and permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 10 µg/mL of Purified Mouse IgG2b, κ Isotype Control (clone MPC-11) (Cat. Description

Von Willebrand factor (VWF) plays a crucial role in blood clotting by assisting platelets in forming clots at the site of blood vessel injury. Acting as a bridge between platelets and the damaged blood vessel walls, VWF facilitates platelet adhesion and aggregation, which are essential steps in the clotting process. Moreover, VWF also stabilizes factor VIII, a key protein in the blood clotting cascade, preventing its degradation and enhancing its availability for clot formation. Mutations or deficiencies in VWF results in defects in platelet adhesion and potentially bleeding disorders such as thrombotic thrombocytopenic purpura, Heyde’s syndrome, and von Willebrand disease. A critical region of VWF is the A2 domain. Contained in this domain is the cleavage site for the ADAMTS13 protease which inactivates VWF.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1 - 10 μg/mL is recommended. For intracellular flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody (clone A21125D/G) is not suitable for use in western blot.

This antibody (clone A21125D/G) is a pool of two monoclonal antibodies, A21125D and A21125G.

For intracellular flow cytometry (ICFC), fixation/permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401) is recommended.
