Total lysates (15 µg protein) from 293T and 293T/TRAF3 knockdown (KD) cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:250 diluted (2 µg/mL) purified anti-TRAF3 antibody, clone 9D7C15 (upper). Proteins were visualized using chemiluminescence detection by incubating with 1:3000 dilution of HRP goat anti-mouse-IgG secondary antibody (Cat. No. 405306). 1:2000 dilution of Direct-Blot™ HRP anti-β-actin Antibody (Cat. No. 643807) was used as a loading control (lower). Lane M: MW ladder.

Description

TNFR-associated factor 3, also known as TRAF3, is a member of the TNF receptor associated factor (TRAF) protein family and was originally identified as an adaptor molecule binding to the cytoplasmic tail of CD40. The engagement of CD40 with its ligand recruits the complex, which consists of TRAF3, TRAF2, and c-IAP1/2. This results in the stabilization of NF-κB inducing kinase (NIK). NIK phosphorylates the IKKα complex and subsequently leads to the activation of the RelB/p52 complex. TRAF3 is involved in the Toll-like receptor (TLR) mediated activation of proinflammatory genes such as type I interferons and IL-10. Pathogens detected by TLR3 and TLR4 recruit the adaptor protein TRIF. In turn, this activates IRF3 via the TRAF3-TBK1-IKKe complex. TRAF3 has been shown to play important roles in the regulation of innate immune response, antibody class switching, and B cell survival.

Antigen Details

Structure
568 amino acids with a predicted molecular weight of 64.5 kD. Contains a C-terminal MATH/TRAF domain responsible for the binding to receptor cytoplasmic domains.
Distribution

Cytoplasm.

Function
TRAF3 mediates the signal transduction from TNF receptors and Toll-like receptors, leading to the activation of NF-κB, MAP kinases, or IRF3. TRAF3 is involved in the regulation of anti-viral response and B-cell survival.
Interaction
TRAF3 forms a homodimer. TRAF3 also forms heterodimers with TRAF2 and TRAF5. TRAF3 also interacts with CD40, TICAM1, TRAFD1, BIRC2, BIRC3, TLR4. TRIP, and TBK1.
Biology Area
Cell Biology, Immunology, Innate Immunity, Signal Transduction
