Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of purified mouse IgG1, k isotype ctrl antibody (Cat. No. 400102) or purified anti-HTRA2 antibody (clone 8G11-4B8-4F10-G5). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a control antibody against a separate epitope of HTRA2. Lane M: Molecular weight marker.

    Whole cell extracts (15 µg total protein) from K562, MOLT-4, Hep G2, and HeLa (positive control) lysates and mouse-brain lysates were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL (1:500 dilution) purified anti-HTRA2 antibody (clone 8G11-4B8-4F10-G5) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker.

Description

Htra2/Omi (high temperature requirement 2) is a mitochondrial serine protease and member of the high-temperature requirement A family of oligomeric serine proteases that functions in the regulation of apoptosis, mitochondrial homeostasis, and protein quality control. Under normal conditions Htra2/Omi is localized to the mitochondria where it serves as a chaperone protein. Under stress conditions Htra2/Omi is released to the cytosol where it mediates apoptosis via proteolytic activity and disruption of caspase-IAP (inhibitors of apoptosis proteins) activity as well as through caspase-independent pathways. Loss or disruption of Htra2/Omi function results in mitochondrial defects and the aggregation of unfolded or misfolded proteins and has been implicated in various neurodegenerative disorders such as Parkinson’s disease, thus making Htra2/Omi an important potential therapeutic target in the treatment of such disorders. Likewise, due to its role as a regulator of cell death, Htra2/Omi may also be an important therapeutic target in the treatment of systemic sepsis and the prevention of Sepsis-Associated Encephalopathy (SAE) as well as a potential oncogenic target and modulator of chemotherapy-induced cytotoxicity.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC on HeLa cells using three different fixation/permeabilization methods: methanol-only, PFA+ methanol, and PFA+ Triton X-100. Staining was only observed with PFA+ Triton X-100 permeabilization and even this was quite faint and only detected at high exposures.

This clone is weakly reactive with mouse Htra2 by western blot.
