Whole cell extracts (15 µg protein) from the indicated tissue lysates were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 1.0 µg/mL (1:500 dilution) of purified anti-CaMKII antibody (clone 6G9) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker.

Description

The Ca2+/calmodulin (CaM)-dependent protein kinases (CaMKs) are multifunctional serine/threonine kinases whose activity are regulated through Ca2+ signaling. CaMKII is expressed as a multimeric protein, which typically is comprised of 12 subunits in physiological conditions. CaMKII is an important member of the calcium/calmodulin-activated protein kinase family. It phosphorylates nearly 40 different proteins, including enzymes, ion channels, kinases, and transcription factors. CaMKII plays critical roles in neural synaptic stimulation and T cell receptor signaling. Recent studies suggested that high levels of CaMKII is also expressed in variety of cancers. It regulates proliferation, differentiation and survival of cancer cells.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.5 µg/ml. For immunohistochemical staining on frozen tisse sections, a concentration range of 2.0 - 10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
