Whole cell extracts (15 µg total protein) from the indicated tissue lysate or cell line were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.5 µg/mL (1:1000 dilution) of purified anti-NOS1 (clone W19138A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat.     IHC staining using purified anti-NOS1 (clone W19138A) on formalin-fixed paraffin-embedded human cerebellum tissue at 10x (left) and 40x (right) objectives. The white box indicates the region of the higher magnification image. The tissue was incubated with 10 μg/mL of the anti-NOS1 antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 Goat anti-rat IgG (red) (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue), and the slide was mounted with ProLong™ Gold Antifade Mountant. Scale bar = 50 μM

Description

Nitric oxide synthase 1 (NOS1) catalyzes the formation of nitric oxide (NO) from L-arginine. NO signals in neurons intracellularly to regulate metabolic activity and dendrite growth. NO also functions in neurons as a neurotransmitter-like signaling molecule, and in muscle cells, where it is a potent relaxer. High concentrations of NO are cytotoxic, so it is utilized by macrophages as a bactericide/tumoricide, and it has also been implicated in the progression of neurodegenerative disorders like Alzheimer’s.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration of 5 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product was tested and failed for IHC-F in mouse brain.
