Whole cell extracts (15 µg total protein) from the indicated cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 μg/mL purified anti-SMARCA2 (BRM) antibody (clone W19134A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat. No. 643807) was used as a loading control at a 1:10,000 dilution (lower). Lane M: Molecular weight marker.

Description

SMARCA2 (also known as BRM) is a subunit of the SWI/SNF complex that facilitates chromatin remodeling. As a catalytic subunit, SMARCA2 contains an ATPase domain which uses ATP to alter DNA-histone interactions within a nucleosome. SMARCA2 as part of the chromatin remodeling complex regulates the expression of genes involved in cell cycle, metabolism, development and cancer. In most cases of cancer there is either downregulation or deficiency of the BRM gene. SMARCA2 serves as a target for anticancer therapy. Mutations in SMARCA2 can result in Nicolaides-Baraitser syndrome.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using 4% PFA-fixed HeLa cells permeabilized with either methanol or Triton X-100. Neither method was compatible with SMARCA2 staining.

This clone failed to enrich SMARCA2 from HeLa extracts when tested for IP.
