Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-RET (clone W22020E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

The proto-oncogene RET encodes a transmembrane tyrosine-protein kinase receptor that is involved in several cellular mechanisms such as proliferation, migration, neuronal navigation, and cell differentiation. RET is important in the development of the nervous system, as well as in the formation of organs and tissues derived from the neural crest. The interaction with the ligand glia cell-line derived neurotrophic factor (GDNF), is vital for promoting cell survival. Conversely, in the absence of GDNF the receptor becomes active and induces apoptosis. Oncogenic activation of RET by either cytogenetic rearrangement or activating mutations, is associated to various diseases such as Hirschsprung disease, central hypoventilation syndrome, and multiple endocrine neoplasia type IIA and type IIB.
