Mouse testicular tissue was formalin-fixed and paraffin-embedded, prior to sectioning. After deparaffinization and antigen retrieval with sodium citrate pH 6, the sample was stained overnight with anti-Ly6k (clone MK34) at 10 µg/ml followed by detection using BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, AEC).

Description

Lymphocyte antigen 6K (Ly6K) is a 17 kD GPI-anchored membrane protein expressed by testicular germ cells. Ly6K associates with TEX101 and may have a role in spermatogenesis. It is also expressed in some cancer cell types; a recent study also demonstrated its expression in plasma cells. A soluble 12 kD form of Ly6K has been reported.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤1.0 µg per million cells in 100 µl volume. For immunohistochemical staining on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Additional reported applications (for the relevant formats) include: immunohistochemical staining on frozen tissue sections1, western blotting1, and immunoprecipitation1.
