Total cell lysates (15 µg total protein) from A431, HaCat, HepG2, HeLa (reduced expression control) and U937(reduced expression control) cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-IGF2BP2 Antibody, clone O93C8, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Actin filaments were labeled with Alexa Fluor® 488 Phalloidin (green). Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

IGF2BP2 belongs to the family of Insulin-like growth factor-II mRNA binding proteins (IMP1, IMP2, and IMP3). Members in this family contain four hnRNP K homology (KH) domains and two RNA recognition motifs (RRM) domains. They are involved in the RNA cytoplasmic localization, stability, and translation. Recent studies suggest that IGF2BP2 and IGF2BP1 bind to target mRNAs via all four KH domains. Studies showed that the expression of IGF2BP2 is associated with liposarcoma, liver cancer, and endometrial adenocarcinomas. Genome-wide association studies have also identified IGF2BP2 as one of the several factors for developing type 2 diabetes.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.5 µg per ml. For immunocytochemistry, a concentration range of 0.1-10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Structure

Distribution

Nucleus, cytoplasm, and the cytoskeleton. It is expressed in oocytes, granulosa cells, Leydig cells, spermatogonia, but is expressed weaker in the lymph nodes, lungs, liver, kidney, heart muscles, spleen, and testis.

Function
IGF2BP2 binds to the 5' UTR of the insulin-like growth factor 2 (IGF2) mRNA and regulates IGF2 translation. It also modulates the cytoplasmic trafficking, turnover, and translation of various target mRNAs.
