Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1 µg/mL Purified anti-Caldesmon (clone W22071A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat.     U-87 MG cells were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 5 µg/mL of Purified anti-Caldesmon (clone W22071A), followed by with Alexa Fluor® 594 Goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Description

Caldesmon is a calmodulin-, actin-, and tropomyosin-binding protein that plays an essential role in smooth muscle and non-muscle contraction. This protein is also involved in cellular mitosis and Schwann cell migration during peripheral nerve regeneration.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For immunohistochemistry (IHC-P), either Citrate Buffer, 10X (Cat. No, 420902) or Tris-EDTA pH 9.0 is recommended for antigen retrieval.

For immunocytochemistry (ICC), fixing with Fixation Buffer (Cat. No. 420801) is recommended. Alternatively, 4% paraformaldehyde fixation followed by either permeabilization with 0.5% Triton X-100 or ice-cold methanol is recommended.

Clone W22071A cross-reacts with mouse in western blotting (WB).

Distribution

Tissue distribution: high-molecular-weight Caldesmon (isoform 1) is predominantly expressed in smooth muscle; low-molecular-weight Caldesmon (isoforms 2, 3, 4 and 5) is widely distributed in non-muscle tissues and cells. Caldesmon is not expressed in skeletal muscle or heart.
Cellular distribution: cytoskeleton, cytosol, and plasma membrane.

Function
Regulates actomyosin interactions in smooth muscle and non-muscle cells which affects cell contractions; involved in cellular mitosis and in Schwann cell migration during peripheral nerve regeneration.
