    IHC staining of purified anti-YY1 (clone W21112E) on formalin-fixed paraffin-embedded human colon. Following antigen retrieval using Tris-EDTA, pH 9.0, the tissue was incubated with 5.0 µg/mL of the primary antibody overnight at 4°C. This was followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) at 2.5 µg/mL. Nuclei were counterstained with DAPI (Cat. No. 422801) (panel A) and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale: 50 µm

Description

Yin Yang 1 (YY1), is a ubiquitously distributed transcription factor that plays a pivotal role in regulating gene expression and coordinating various cellular processes. By interacting with other transcription factors, co-regulators, and epigenetic modifiers, YY1 can modulate gene expression to control diverse biological processes, including cell proliferation, differentiation, apoptosis, and development. Based on its interactions with other proteins such as MYC, YY1 can function as a transcriptional activator or repressor. Furthermore, YY1 is involved in maintaining genomic stability and has been implicated in DNA repair mechanisms. Its ability to interact with a wide range of regulatory elements and its context-dependent transcriptional activities make YY1 an essential factor in orchestrating intricate gene regulatory networks within the cell, and a potential target for cancer therapies.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone is not recommended for ICC application.

For IHC-P antigen retrieval with either Tris-EDTA, pH 6.0, or sodium citrate, pH 9.0 (e.g. Cat. No. 420901) is recommended.
