    SR cells untreated (panel B) or treated with TBB (CK2 inhibitor) (panel C) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801), permeabilized with cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then intracellularly stained with 5 µg/mL of Purified anti-PTEN Phospho (Ser380) (clone A22005D) (panels B and C) or Mouse IgG1, κ Isotype Control (Cat. No. 401401) (panel A), followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured in a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm

Description

PTEN is a tumor suppressor gene which encodes a phosphatase that preferentially dephosphorylates phosphoinositide substrates and thereby modulates cell cycle progression and cell survival. PTEN is an important negative regulator of the PI3K/AKT/mTOR signaling pathway by dephosphorylating PIP3. PTEN is commonly mutated in human cancer, hence, loss of expression/function of PTEN due to mutations have been observed in prostate cancer, glioblastoma, endometrial, lung and breast cancers. Other non-cancerous clinical disorders associated with PTEN mutation are Cowden syndrome, Bannayan-Riley-Ruvalcaba syndrome, and proteus-like syndrome. PTEN is phosphorylated at several residues and some lead to increase in activity while others lead to loss of function and degradation. Specifically, phosphorylation at serine 380 residue by casein kinase 2 (CK2), leads to degradation of PTEN in the proteasome.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 2.5 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

ICC: This antibody is compatible with either PFA/Triton X-100 or PFA- Methanol fixation/permeabilization protocols. It is not suitable to use on methanol only fixation/permeabilization protocols.

ICFC: This antibody is suitable to use in cells fixed with either Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) or True-Phos™ Perm Buffer (Cat. No. 425401) systems. Not tested for use with the True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401).
