Whole cell lysates (15 µg) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with 1.0 µg/mL (1:500 dilution) purified anti-MUT antibody (clone W18259A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker.

Description

MUT encodes MCM (Methylmalonyl-CoA mutase), which resides in the mitochondria to metabolize branched and odd-chain fatty acids. Utilizing cobalamin as a cofactor, MCM also isomerizes methylmalonyl-CoA to make succinyl-CoA, an important intermediate in the Kreb’s Cycle. Mutations in the gene can lead to disruptions in amino acid metabolism and may alter lipid and glucose homeostasis.

Application

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.2 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

When tested for ICC on 4% PFA-fixed HeLa cells, this clone produced punctate nuclear staining, which is inconsistent with the reported mitochondrial matrix localization of MUT. We therefore do not recommend this clone for the application.
