IHC staining of purified anti-CRMP5 (clone A18231D) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using Tris-EDTA pH 9.0 buffer (10 mM Tris, 1 mM EDTA), the tissue was incubated with (panel A) and without (panel B) 5.0 µg/mL of purified anti-CRMP5 (clone A18231D), followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 20 µm

    Whole cell extracts (15 µg total protein) from the indicated cell line treated with siRNA control (-) or CRMP5 siRNA (+) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL of purified anti-CRMP5 (clone A18231D) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

CRMP5 (Collapsin Response Mediator Protein 5 or DPYSL5) is a member of the CRMP family and is involved in regulation of the cytoskeleton. CRMP5 acts as a negative regulator of microtubule stability and dynamics and participates in the fibroblast growth factor (FGF) signaling pathway, regulating axon guidance and neuronal development. CRMP5 has been implicated in several human diseases. In cancer cells, CRMP5 regulates cancer cell migration, invasion, and proliferation through its interaction with the tumor suppressor protein, PTEN. Mutations in CRMP5 are related to neurodevelopmental disorders, including X-linked intellectual disability and autism spectrum disorder. CRMP5 regulates tau protein degradation through its interaction with microtubules and enzymes, suggesting its role in neuroinflammation and Alzheimer's disease.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product is not recommended for use in immunocytochemistry (ICC).

For use in immunohistochemistry (IHC), antigen retrieval with Tris-EDTA pH 9.0 is recommended.
