CD44+ cells were clearly distinguished from CD44- mouse peripheral blood leukocytes.

Experimental Design

Application:	Flow cytometry
Cells used:	Mouse peripheral blood leukocytes
Brief Protocol:	Collected 25 uL blood. Washed once in cold PBS. Lysed red blood cells. Washed once in cold PBS. Stained cells in 100 uL cold PBS containing Live/Dead and TruStain Fc block (2 ug/mL) on ice for 15 minutes. Added 100 uL cold PBS containing isotype control (4 ug/mL) or CD44 PE (4 ug/mL) and incubated on ice for 15 minutes. Washed in cold PBS. Fixed in 100 uL 1.5% PFA until analysis.
Results Summary:	CD44+ cells were clearly distinguished from CD44- mouse peripheral blood leukocytes.
Additional Notes:	All plots show live single cells based on live/dead exclusion. Left contour plot shows staining of peripheral blood leukocytes with isotype control. Middle contour plot shows staining of peripheral blood leukocytes with CD44 PE. Right contour plot shows overlay of peripheral blood leukocytes stained with isotype control (red) or CD44 PE (black).
