Purified CD38 was conjugated to 148Nd and worked well in a mass cytometry panel.

Experimental Design

Application:	Mass cytometry
Cells used:	Human PBMC
Brief Protocol:	We conjugated purified CD38 to 148Nd using a MaxPar® Antibody Labeling Kit (we followed the kit instructions exactly). We then used our conjugated CD38-148Nd in a mass cytometry panel to characterize human PBMCs. We looked at CD38 expression with and without SEB stimulation and found that on the CyTOF® we detected comparable frequencies of CD38+ T and B cells as we did with fluorescence cytometry (R=0.87, p=0.0002).
Results Summary:	We conjugated CD38 to 148Nd and used it in a mass cytometry panel. It detected the same frequency of CD40L+ singlets as fluorescence cytometry did (using CD40L-PE) in 6 subjects (R=0.87, p=0.0002).
