Total protein from HeLa (negative control cell line), K562 and NIH3T3 cell lysates (15 µg protein) were resolved by 4-12% Bis-tris gel electrophoresis, transferred to nitrocellulose, and probed with 1 µg/mL purified anti-EOS antibody (clone W15169A, upper). Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (lower).

Description

EOS, also known as IKZF4 (Ikaros family zinc finger 4), is a member of the Ikaros family of zinc finger transcription factors, which are key transcriptional repressors during development of lymphocytes. Each of the five Ikaros family members (Ikaros, Helios, Aiolos, Eos and Pegasus) has N-terminal and C-terminal zinc-finger motifs responsible for DNA binding and dimerization, respectively. EOS is a highly conserved DNA-binding protein that recognizes the typical 5'WGGAAT-3' Ikaros-binding sequence. EOS has been implicated in the development of lymphoid cells and nervous systems, as well as mediating FOXP3-dependent gene silencing in CD4+ regulatory T cells though interacting with FOXP3 and CTBP1.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1:1000 - 1:250 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone W15169A does not cross-react with mouse (in-house tested). This antibody recognizes both isoforms of EOS and it also detects a non-specific protein of approximately 75 kD in the K562 cell line tested.
