Jurkat cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for three minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with 1 µg/ml Alexa Fluor® 594 anti-human TAL1 (clone 4G8C07) antibody (red) overnight at 4°C. Nuclei were counterstained with DAPI (blue). The image was captured by a 60X objective.

Description

TAL1, also known as SCL, is a class I basic helix-loop-helix (bHLH) transcription factor. TAL1 is essential for the establishment of hematopoietic stem cells during embryogenesis, and regulating proliferation and differentiation of multiple hematopoeitic cell lineages, such as erythrocytes, megakaryocytes, mast cells, and eosinoohils. Tal1-null mice are embryonic lethal due to the failure of yolk sac hematopoiesis and vascular defects. In hematopoietic cells, TAL1 forms a multiprotein complex with other transcription factors, which include the E protein TCF3, LMO2, LDB1, and GATA family members. The large protein complex containing TAL1 binds to specific cis-regulatory elements encoding the consensus sequence AACAGATGGT and regulates expression of target genes. TAL1 is not expressed in normal lymphoid cells, but ectopic expression of TAL1 in T-cells leads to T-cell acute lymphocytic leukemia (T-ALL).
