Whole cell extracts (15 µg total protein) from the indicated cell lines were boiled at 100°C for 5 minutes and were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 μg/mL of purified anti-SMC3 (clone DJ5.2.3) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Lane M: Molecular weight marker

Description

Structural maintenance of chromosomes protein 3 (SMC3) is a member of the SMC protein family and a component of the cohesin complex. Cohesin plays an important role in mitosis and meiosis by mediating sister chromatid cohesion, homologous recombination, and spindle pole formation. SMC3 activity is mediated by phosphorylation at a pair of conserved lysine residues, stabilizing the cohesion complex during mitosis. Mutations in SMC3 can result in Cornelia de Lange syndrome, characterized by developmental disabilities.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Product was tested for use in ICC and IHC-P applications but did not meet passing criteria.
