IHC staining of Purified anti-Neu1 (clone A18167C) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0), the tissue was incubated without (panel A) and with (panel B) 10 µg/mL of Purified anti-Neu1 (clone A18167C) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 50 µm

Description

Neu1, a member of the sialidase enzyme family, plays a crucial role in cellular processes by removing sialic acid (N-acetylneuraminic acid) residues from glycoproteins and glycolipids on the cell surface. This process, known as desialylation, impacts various cellular functions, including cell-cell interactions, signal transduction, and immune responses. Additionally, desialylation recycles sialic acid, contributing to the maintenance of cellular sialic acid levels critical for normal physiological processes. Alterations in Neu1 activity leads to sialidosis, a lysosomal storage disease resulting in an accumulation of sugar-protein complexes.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody (clone A18167C) is not suitable for Western blot (WB) or immunocytochemistry (ICC).

For immunohistochemistry (IHC), we recommend antigen retrieval with Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0). Antigen retrieval with Sodium Citrate is not recommended.
