    Whole cell extracts (250 µg total protein) prepared from HEK293T cells treated with 100 µM DFO overnight were immunoprecipitated overnight with 2.0 µg of purified rat IgG2a, κ isotype control (Cat. No. 400502) or purified anti-IREB2 (clone W18260A). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with HRP Donkey anti-rabbit IgG (clone Poly4064) (Cat.     IHC staining of purified anti-IREB2 (clone W18260A) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated without (panel A) and with (panel B) 5 μg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 50 μm

Description

IREB2 is an iron-responsive protein that regulates a cell’s levels of iron, a key micronutrient. In low-iron situations, IREB2 is free to bind to iron-responsive elements in mRNA molecules, stabilizing transferrin mRNA to increase expression of the iron uptake receptor, while blocking the translation of ferritin mRNA, a protein which sequesters excess iron molecules. However, when sufficient iron levels are present, IREB2 undergoes a conformation change to lose its mRNA binding ability, resulting in the degradation of transferrin mRNA while releasing the translation block of ferritin. Variants in the gene encoding IRP2 have been associated with lung cancer and chronic obstructive pulmonary disease.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product was tested for use in ICC using PFA-Triton-X, PFA-MeOH, and MeOH only fix-perm methods, and ICFC using the Cyto-fix method, but did not meet passing criteria. 

This product was tested for mouse reactivity in WB and ICFC but did not meet passing criteria.
